Design and delivery of messenger RNA-based vaccines

چکیده
In these last years, we are witnessing the emergence of a new class biopharmaceuticals based on messenger RNA (mRNA). One most promising applications mRNA is its use as vaccines. Many reports, including ours, have demonstrated preclinical efficacy vaccines several advantages over traditional or even DNA Unlike attenuated inactivated vaccines, encodes for specific antigen that will be expressed in situ and stimulates both innate immune system an adaptive immunity to promote humoral cellular responses.

Nonviral delivery of self-amplifying RNA vaccines.
Despite more than two decades of research and development on nucleic acid vaccines, there is still no commercial product for human use. Taking advantage of the recent innovations in systemic delivery of short interfering RNA (siRNA) using lipid nanoparticles (LNPs), we developed a self-amplifying RNA vaccine. Here we show that nonviral delivery of a 9-kb self-amplifying RNA encapsulated within ...

Genetic modification of dendritic cells with plasmid DNA is plagued with low transfection efficiencies because DNA taken up by non-dividing dendritic cells rarely reaches the nucleus. But this difficulty can be overcome by the use of messenger RNA (mRNA), which exerts its biological function in the cytoplasm and obviates the need to enter the nucleus. Since pioneering work of Boczkwoski et al.,...
متن کاملSublingual Delivery of Vaccines
Sublingual immunization, where the vaccine is placed directly under the tongue and absorbed through the sublingual mucosa, has the advantages of ease of delivery without the complication of oral administration or the safety concerns of intranasal vaccines. Sublingual vaccine delivery has been shown to induce development of humoral and cell mediated immunity in both the systemic and mucosal comp...
متن کاملMessenger RNA Modulated by Thioacetamide Increased Activity of Rat Liver Messenger RNA and of Albumin
Administration of thioacetamide to rats was found to increase the activity of liver messenger RNA in the wheat germ protein synthesis assay. The synthesis of total protein was increased about 2.5-fold after 4 days of treatment. The treatment was associated with an increase in the relative quantity of polyadenylic acid-containing RNA. Immunoprecipitation studies showed that albumin synthesis dir...
